摘要
目的 探明 Bcl- x基因在大鼠胰岛细胞凋亡时剪切方式的变化及其该基因在胰岛细胞凋亡中的作用。方法 应用 TUNEL法检测长期高糖培养时大鼠胰岛细胞凋亡 ,应用半定量多重 RT- PCR(SQ- RT- PCR)检测了胰岛细胞凋亡时 Bcl- x基因 m RNA的表达变化情况 ;同时观察小剂量 STZ所致的糖尿病大鼠胰岛 Bcl- x基因 m RNA的表达变化。结果 2 5 .5 m M葡萄糖浓度体外培养 14天较 5 .5 m M和 11.1m M葡萄糖浓度培养 ,胰岛细胞凋亡百分率明显增加 ,分别为 31.5± 4 .5 % ,15 .4± 3.0 % ,17.4± 4 .8% ;P <0 .0 1。细胞凋亡百分率变化的同时伴有 Bcl- x基因转录后剪切方式的改变 ,表现为 Bcl- x S m RNA表达明显增加 ;低剂量 STZ所致的糖尿病大鼠胰岛 Bcl- x S m RNA表达明显增加 ,Bcl-x L与 Bcl- x S比率明显减少。结论 长期高糖可以诱导大鼠胰岛细胞凋亡 ,Bcl- x基因剪切方式的改变所致的 Bcl- x L/Bcl- x S比率的变化在高糖和
Objective To observe the change of expression of Bcl x in rat islets underwent apoptosis and investigate effects of splicing of transcribed Bcl xL gene on apoptosis of rat islet cells.Methods Apoptotic islet cells exposed to sustained high glucose were identified by TUNEL method. Using semi quantitative multiplex RT PCR the expression of Bcl x was measured in rat islet cell and STZ induced diabetic islet. Results The proportion of apoptotic rat islet cells exposed to 25 5mM glucose was significantly higher than those of islet cells cultured with low 5 5mM and 11 1mM glucose, 31 5±4 5%,15 4±3 0%,17 4±4 8% respectively, P<0 01. Expression of Bcl xS mRNA increased 14 days after incubation with 25 5mM glucose and the ratio of Bcl xL/Bcl xS decreased in rat islet cell. Those changes of the expression of Bcl x were also observed in STZ induced diabetic rat islet. Conclusion Sustained high glucose concentration can induce apoptosis of islet cell. The increase of the expression of Bcl xS in STZ induced diabetic rat islet and islet cell cultured with sustained high glucose suggest that the change of splicing of transcribed Bcl x may be play a role in apoptosis of islet cell. Furthermore the ratio of Bcl xL/Bcl xS may be an important predictor of islet cell apoptosis.
