摘要
目的 :研究体外转染canstatin核糖核酸 (RNA)对培养的兔血管平滑肌细胞 (VSMC)凋亡的影响。方法 :在无菌条件下取新西兰白兔胸主动脉 ,采用阳离子脂质体介导的基因转染方法 ,将外源性canstatinRNA转入VSMC中 ,应用流式细胞仪 (FCM )、TUNEL及电镜观察canstatinRNA转染细胞的凋亡。实验共分对照组、DOSPER组和canstatin组。结果 :阳离子脂质体介导的canstatinRNA成功转入VSMC并表达 ,流式细胞仪检测表明 ,canstatinRNA转染细胞的细胞周期发生细胞生长期 (G1期 )阻滞 ,G1期细胞 ( 5 8 93± 2 17) %较对照组 ( 4 9 13± 2 0 7) %及DOSPER组 ( 4 9 99± 1 45 ) %增加 ,有显著性差异 (P <0 0 5 ) ,脱氧核糖核酸合成期 (S期 )细胞 ( 19 88± 2 2 8) %较对照组 ( 2 8 0 6± 3 77) %及DOSPER组( 2 8 5 0± 4 41) %减少 ,有显著性差异 (P <0 0 5 )。canstatin组RNA转染细胞的凋亡率 ( 17 74± 3 93) %高于对照组 ( 4 89± 3 5 9) %及DOSPER组 ( 4 0 6± 1 80 ) % ,有极显著性差异 (P <0 0 1) ,而对照组及DOSPER组无显著性差异 (P >0 0 5 )。TUNEL检测显示 ,canstatin基因转染细胞凋亡明显增加 ,电镜下观察到转染细胞出现典型凋亡特征性改变。结论 :阳离子脂质体介导的canstatinRNA成功转染VSMC并能显著?
Objective:To investigate the effects of exogenous canstatin RNA transfection on apoptosis of cultured rabbit vascular smooth muscle cells(VSMC)in vitro. Methods:By means of cationic liposome(DOSPER)mediated method,canstatin RNA was transfected into cultured VSMC.Cell cycles and apoptosis were detected by FCM,TUNEL and electromicroscope. Results:Canstatin RNA was effectively transfected into cultured primary rabbit smooth muscle cells by the cationic liposome(DOSPER) and was successfully expressed.In canstatin RNA transfected cells,cycle arrested in G 1 phase (58.93±2.17)%,(49.13±2.07)% in the control group and(49.99±1.45)% in the DOSPER group,(p<0.05).The percentage of canstatin RNA transfected cells in S stage was decreased markedly compared with that of the control group and DOSPER group.(19.88±2.28)% vs.(28.06±3.77)% and(28.50±4.41)%,respectively,(p<0.05).The percentage of apoptosis in canstatin RNA transfected cells was increased significantly compared with that of the control group and DOSPER group.(17.74±3.93)% vs.(4.89±3.59)%and(4.06±1.80)%,(p<0.01).The effect of canstatin RNA-induced apoptosis was further confirmed by TUNEL and electromicroscopy. Conclusion:Canstatin RNA can be effectively transfected into cultured rabbit VSMC by the cationic liposome(DOSPER).The expression of canstatin RNA.
