摘要
目的 观察NGF GDNF基因修饰神经干细胞 (NSC)单独和联合移植对AD模型鼠的学习记忆改善作用。 方法 单侧切断SD大鼠穹窿海马伞 (FF)制备AD模型鼠。术后 8~ 10d ,侧脑室移植基因修饰及未修饰的NSC。移植后 2周 ,进行Morris水迷宫行为学检测。 结果 NGF组和NGF +GDNF组的后 3个时间段的平均逃避潜伏期较损伤组和NSC组明显缩短 (P <0 0 1) ;平台象限游泳距离百分比明显增高 (P <0 0 1) ;GDNF组的平台象限游泳距离百分比高于NSC组 (P <0 0 1) ,但平均逃避潜伏期与NSC组无明显差异。 结论 NGF GDNF基因修饰NSC单独和联合移植对AD模型鼠的行为学有不同程度的疗效 ,其中NGF组和NGF +GDNF组疗效较好 ,但后者并未优于前者 。
Objective To investigate the memory amelioration of the AD model rat after a single or combined transplantation of the neural stem cells(NSCs) modified with gene of NGF or GDNF. Methods The AD model rat was made by cutting unilaterally the fibria\|fornix of male SD rat.Eight to ten days after surgery,the genetically modified and unmodified NSCs were implanted into the lateral cerebral ventricle of the rats.Two weeks after transplantation,the amelioration of memory impairment of the rats were detected by Morris water maze. Results The average escape latencies of the last three blocks in NGF and NGF+GDNF groups were lesser,and the percentages of swim distance in the platform quadrant were greater than that in NSC group( P <0\^01).The percentage of swim distance in the platform quadrant in GDNF group was also greater than that in NSC group( P <0 01),but there was no significant difference in the average escape latency between them. Conclusion\ The behavioral amelioration of AD model rat was obtained in different extent by a single or combined transplantation of the NSCs modified with gene of NGF or GDNF.The effect in NGF and NGF+GDNF groups was better than that in GDNF group,but there was no better therapcutic effect was observed in NGF+GDNF group than that in NGF group.\;[
