摘要
用计时电流法成功地研究了尼古丁对乳酸脱氢酶活性的影响 ,测定了尼古丁存在与否时乳酸脱氢酶酶促反应的初速度V0 、酶促反应最大反应初速度Vm 及米氏常数Km 。实验结果表明尼古丁对乳酸脱氢酶的活性有很大影响 ,但在高浓度的NADH存在下 。
Chronoamperometry based on the 'controlling-diffusion layer' concept of the convective system was used to monitor the effect of nicotine on the activity of lactate dehydrogenase (LDH) at a bare glassy carbon (GC) electrode. The real initial reaction rate (V-0) and the maximum reaction rate (V-max) of the enzyme-catalyzed reaction and the Michaelis-Menten constant (K-m) were determined. Analysis of the experimental results revealed that the nicotine ion could inhibit the LDH activity remarkably. But, when the concentration of nicotinamide adenine dinucleotide (NADH) was high, the inhibition would be decreased deeply.
