瓜蒌皮用不同浓度乙醇回流提取 ,拮抗实验表明 ,70 %乙醇提取物钙拮抗作用最强 ;将70 %乙醇提取物用石油醚萃取 ,萃取物经硅胶层析后的各组分 ,用4 5Ca跨膜测量方法研究其对大白兔主动脉PDC钙通道Ca2 + 内流的影响 ,以探明瓜蒌皮治疗冠心病的有效成分。结果表明 ,组分 1、4、5具有明显的钙拮抗作用 ,且组分 5活性最好 ;组分
The active fractions of fructus trichosanthes cortex with antagonistic effect were studied by 45 Ca isotopic tracer technique- 45 Ca transmembrane influx technique. The fructus trichosanthes cortex was first extracted by 30-100% ethyl alcohol. The calcium antagonism of the extract obtained by 70% ethyl alcohol was found to be the highest. This extract was further extracted by petroleum ether, then isolated on Silica gel columm chromatography with elution of ethyl acetate and 95% ethyl alcohol. It was found that the eluates have five fractions. Among them, the fifth fraction had the strongest Ca 2+ antagonistic effect. In contrast, the third fraction has obvious activiating effect, but no antagonistic effect.
