切断部分背根对脊髓Ⅱ板层c-jun,c-fos表达的影响——免疫组织化学研究 被引量:3

摘要 目的 探讨 c- fos、c- jun与脊髓可塑性的关系。方法 将成年雄猫 2 5只分为正常组、假手术后 3天组 ,假手术后 10天组、单侧备用根术 (切除一侧 L1 - L5,L7- S2 背根节 ,保留 L6为备用根 )后 3天组和单侧备用根术后 10天组 (每组 5只 )。取各组动物的 L5脊髓用 4%多聚甲醛固定后制作 2 0μm厚冰冻切片。每例动物间隔取 5张切片 ,分别用 c- fos (稀释度 1∶ 30 0 0 ,Sigm a) ,c- jun (稀释度 1∶ 10 0 0 ,Santa Cruz)抗体行 ABC免疫组化染色。 DAB棕色反应显色。结果 正常组及假手术组猫脊髓灰质各层内 ,c- jun的免疫阳性反应物主要出现在神经元胞核 ,但胞浆亦有弱阳性反应。部份去背根后 3天及 10天 ,单侧备用根手术组 板层 c- jun阳性神经元的数量及反应强度均较正常组及假手术者明显增多 (P<0 .0 5 )。但术后 3天与 10天组间无显著差异 (P>0 .0 5 )。与 c- jun比较 ,各组脊髓内均未见 c- fos的阳性细胞。结论 切断部分背根导致脊髓 板层 c- jun表达上调 ,提示 c- jun可能与脊髓 板层可塑性有关。 ? Objective To investigate the relationship between c fos, c jun and the plasticity of spinal lamina Ⅱ. Method Adult cats(n=25) were divided into a normal group, 2 sham operation groups (3rd and 10th day after operation) as well as 2 experimental groups (3rd and 10th day after unilateral partial dorsal root rhizotomy, where L 1 L 5 and L 7 S 2 DRG were resected, but L 6 was spared). The animals were sacrificed and the segments of L 5 spinal cords were taken and sectioned into frozen sections of 20 μm in thickness. Five sections of each animal were stained with specific antiserums against c fos and c jun (Santa Cruz, 1∶1000) using immmunohistochemical ABC method. The number and stain intensity of both c fos and c jun positive neurons in spinal lamina Ⅱ were counted and analyzed respectively. Result c jun positive reactive products were mainly distributed in the neuronal nucleus, but the neuronal cytoplasm was also weakly stained in the normal and sham operation groups. Both the number of positive neurons and the stain intensity of c jun in lamina Ⅱ( L 5 ) increased apparently at day 3 and day 10 after partial dorsal root rhizotomy compared with those of the normal and sham operation groups( P <0. 05). Howerer, no c fos positive neuron in spinal lamina Ⅱ was observed in any group. Conclusion The results indicate that c jun might be related to the plasticity of spinal lamina Ⅱ following partial dorsal root rhizotomy. The experiment of c jun in lamina Ⅱ was up regulated following partial dorsal root rhizotomy. \;〔
