马桑内酯慢性致痫大鼠海马星形胶质细胞的激活 被引量:3

摘要 目的 研究慢性癫痛大鼠点燃时海马星形胶细胞的激活情况。方法 采用马桑内酯慢性癫痫大鼠模型 ,观察大鼠点燃后海马 NF-κBp6 5和胶质原纤维酸性蛋白 (glial fibrillary acidic protein,GFAP)免疫细胞化学反应(im munoreactivity,IR)的变化。结果 点燃后 1h,海马 CA1区 GFAP- IR开始增强 ,4~ 8h可观察到 GFAP- IR阳性细胞数量增多并明显浓染 ,这种强 GFAP- IR持续至点燃后 2 4h;点燃后 1h,NF- κBp6 5即可在海马 CA1区神经元和胶质细胞内表达 ,主要位于胞核内 ,至 8h阳性神经元细胞核基本消失而可见大量 NF- κBp6 5 - IR阳性的胶质细胞 ;双重免疫细胞化学方法显示 GFAP/ NF- κBp6 5 - IR阳性细胞在点燃后 1h即可观察到 ,4h达最高峰 ,2 4h恢复至对照水平。结论 马桑内酯慢性致痫大鼠点燃时星形胶质细胞表现出一种早期而持续的激活 ,提示反复激活的星形胶质细胞对癫痫的复发可能起重要的作用。 ? Objective To discover the condition of activation of hippocampal astrocytes after seizure chronic epilepsy rats. Method The expression of NF κBp65 and glial fibrillary acidic protein(GFAP) in the hippocampus of chronic epilepsy rats kindled by systemic use of Coriaria Lactone was studied by immunohistochemical methods. Result The GFAP immunoreactivity (IR) began to increase at 1h after seizure.Obvious increase in the number and staining intensity of GFAP IR positive cells was found at 4 to 8h,and this high GFAP IR remained to 24h after seizure. The NF κBp65 expression in neurons and neuroglia of the CA1 area of hippocampus,mainly in the nucleus, was induced at 1h.At 8h after seizure,the NF κBp65 IR positive neurons disappeared, while a lot of NF κBp65 IR positive neuroglia could be observed. Double immunocytochemical staining showed that the GFAP/ NF κBp65 IR positive cells appeared at 1h, reached the top level at 4h and returned to control level at 24h. Conclusion The rapid and sustained increase in the NF κBp65 IR of astrocytes after seizure in our model indicates that the repeatedly activated astrocytes might play an important role in the relapse of epilepsy. \;〔
