银杏叶提取物对大鼠肝星状细胞增殖的影响及机制研究 被引量:3

摘要 目的研究肝纤维化过程中银杏叶提取物(GbE)抑制肝星状细胞(HSC)增殖的机制。方法用不同浓度GbE(0、50、100、200、300 mg/L)处理HSC 24 h后,MTT法检测GbE对HSC-T6增殖的影响,Western blotting和RT-PCR法测定GbE对P21/P27蛋白和mRNA表达的影响。结果 MTT检测结果显示,GbE在100 mg/L时,抑制HSC-T6的增殖(P<0.05),浓度为200和300 mg/L时,抑制增殖作用明显增强(P<0.01);Western blotting检测结果显示,GbE明显增加P21/P27蛋白的表达,且促进P27蛋白表达的作强于P21;进一步用RT-PCR检测发现,GbE也能增加P21/P27 mRNA的表达。结论 GbE能够抑制HSC-T6的增殖,这种增殖抑制作用主要是通过增加P21/P27蛋白和mRNA的表达实现的。 Objective To investigate the mechanism of ginkgo biloba extract(GbE)in inhibiting the proliferation of hepatic stellate cell(HSC-T6)during the process of hepatic fibrosis. Methods HSC-T6 was cultured with GbE at the concentration of 0,50, 100,200 and 300 mg/ L for 24 hours;MTT colorimetric assay was applied to detect the proliferation of HSCs;Western blotting and RT-PCR were applied to observe the effect of GbE on the expression of P21 / P27 in protein and mRNA levels. Results The results of MTT colorimetric assay showed GbE inhibited the proliferation of HSC-T6 at the concentration of 100 mg/ L(P ﹤ 0. 05),while the growth inhibition of GbE on HSC-T6 became more effective at the concentration of 200 and 300 mg/ L( P ﹤ 0. 01);the result of Western blotting showed GbE up-regulated the expression of P21 and P27 in protein and mRNA levels and the up-regulation was more effective on P27. Conclusions GbE can inhibit the proliferation of HSC-T6 through increasing the expression of P21 / P27 protein and mRNA.
