摘要
目的 建立简便、灵敏、试剂稳定性好的分光光度法测定血清镁。方法 在表面活性剂TritonX 1 0 0存在下 ,用国产常用试剂偶氮氯膦Ⅰ作显色剂 ,EGTA掩蔽钙 ,在pH 9 8三羟甲基氨基甲烷(Tris)缓冲体系中分光光度法测定血清镁。结果 该方法显色络合物最大吸收波长为 582nm ,线性范围达 3 0mmol/L ,表观摩尔吸光数为 1 98× 1 0 4 L/ (mol·cm)。回收率为 99 4%~ 1 0 2 2 % ,批内变异系数 (CV)和批间变异系数 (CV)分别为 3 2 %与 3 8% ,与甲基百里酚蓝 (MTB)光度法比较具有良好的相关性 ,Y =0 989X +0 0 1 1 ,r =0 981 4 ,P >0 0 5 ,67名健康人血清镁含量为 0 70~ 1 1 5mmol/L( x±2s)。结论 该法具有快速、简便、灵敏可靠等优点 。
Objective To establish a simple, sinsitive and stable method of determination of magnesium in serum by UV Spectrophotometry Methods Chlophosphonazo Ⅰwas used as a coloring reagent to determine magnesium by spectrophotometry in the presence of surfactant Triton X 100 Results The maximum absorption of the complex compound was at 582 nm, The linearity was 0~3 0 mmol/L The molar absorption coefficient is 1 98×10 4 L/(mol·cm) The recoveries ranged from 99 4% to 102 2% The withinrun and betweenrun CVs are 3 2% and 3 8% Comparing this method ( Y ) with MTB ( X ), we obtained the regression equation Y =0 989 X +0 011, r =0 981 4, P >0 05, The reference inteval ( ±2s ) was 0 70~1 15 mmol/L ( n =67) Conclusions This method is simple, sensitive, rapid and accurate,and suitable for the routine manual or automatic analysis of magnesium in serum
