摘要
目的 探讨孕兔长期使用可卡因对新生兔肾脏的影响。 方法 2 4只孕兔随机分为二组 ,观察组孕兔静脉注射可卡因 [5 m g/ (kg· d) ],对照组注射生理盐水 [1ml/ (kg· d) ],每日一次 ,实验自兔怀孕 15 d开始至分娩为止。收集 15 3只新生兔血、尿标本 ,取肾脏作组织病理学检查。 结果 (1)肾脏光镜下观察无明显异常。电镜下观察组肾小球毛细血管上皮细胞足突融合、线粒体和滑面内质网肿胀 ,肾小管上皮细胞肿胀。 (2 )血尿素氮和肌酐两组差异无显著性 (P>0 .0 5 ) ,血和尿微量球蛋白在观察组分别为 (0 .0 90± 0 .0 30 ) mg/ L、(0 .0 90± 0 .0 2 0 ) m g/ L ,较对照组的 (0 .0 40± 0 .0 10 )mg/ L、(0 .0 5 0± 0 .0 2 0 ) m g/ L显著增高 (P<0 .0 1) ;尿 N-乙酰 -β- D氨基葡萄糖苷酶在观察组为 (9.6± 2 .8) U/ L ,显著高于对照组 (1.9± 0 .6 ) U / L (P<0 .0 1) ;尿表皮生长因子观察组为 (2 .4± 0 .8) m g/L ,对照组为 (3.3± 1.2 ) m g/ L (P<0 .0 5 )。 结论 孕兔长期使用可卡因将对新生兔肾脏造成损害。血、尿β2 - MG和尿 N-乙酰 -β- D氨基葡萄糖苷酶的检测为肾损害的敏感指标。
Objective To explore the effect of chronic administration of cocaine in pregnant rabbits on the kidney of neonatal rabbits. Methods Twenty four pregnant rabbits were randomly divided into two groups. Twelve rabbits received an aqueous solution of cocaine hydrochloride [5 mg/(kg·d).i.v] daily since the fifteenth day in pregnancy, while 12 control pregnant rabbits were injected by a saline solution (i.v.).The serum and urine samples from neonatal rabbits were collected after delivery. The renal histopathological study and the function examination were performed. Results 1.The histopathological study of the kidney showed damage to epithelial cell projecting and swelling of mitochondria and smooth endoplasmic reticulum in transmission electron microscope. 2.There were no significant difference in serum BUN and Cr between two groups(P>0.05). The level of serum β 2-MG and urineβ 2-MG in the cocaine users(0.090±0.030)mg/L、(0.090±0.020)mg/L respectively were significantly higher than that in the controls(0.040±0.010) mg/L、(0.050±0.020)mg/L (P<0.01). The level of urine NAG enzyme in cocaine group (9.6±2.8)U/L was significantly higher than that in the controls(1.9±0.6)U/L (P<0.01). Urine EGF was significantly decreased in cocaine group (2.4±0.8) mg/L than that in control(3.3±1.2)mg/L(P<0.05). Conclusion The results suggest that the kidney of neonatal rabbits were damaged by chronic administration of cocaine. Examination of level of serum, urine β 2-MG, urine NAG enzyme and urine EGF have important significance to renal lesion.
