腺病毒介导的含第一内含子pGH cDNA在CHO细胞中的表达 被引量:1

摘要 通过同源重组的方法分别构建了在CMV启动子控制下的带有第一内含子和不带内含子的pGHcDNA基因的重组腺病毒。通过感染CHO细胞表明 ,构建的重组腺病毒能够介导pGH基因在CHO细胞中表达 ,加入第一内含子的pGHcDNA基因的表达效率比不加内含子的提高了 117% ,这说明pGH基因的第一内含子有促进该基因在真核细胞中表达的功能。 The recombinant adenoviruses containing pGH cDNA and pGH cDNA with the first intron under the control of CMV promoter were constructed respectively by homogenous recombination method.The results showed that the recombinant adenovi ruses could mediate pGH cDNA expression in CHO cells infected with the recombinant adenoviruses.The expression level of pGH cDNA with the first intron increased by 117% compared with pGH cDNA without intron.This indicate that the first intron of pGH gene have the function of improving the expression of the pGH gene.
