摘要
以热板法镇痛效应为指标,测定了川芎挥发油表观药动学参数。给药剂量分别为ip52mg/kg、96mg/kg。药效动力学参数腹腔给药为:Ke=0.1138h1,Ka=3.3261h1,t1/2(ke)=6.09h,t1/2(ka)=0.21h,T(peak)=1.05h,C(max)=50.6255mg/kg,AUC=526.2315(mg/kg)·h,CL/F(s)=0.1637mg/kg/h/(mg/kg),V/F(c)=0.9114(mg/kg)/(mg/kg);灌胃给药为:Ke=0.0929h1,Ka=0.7693h1,t1/2(ke)=7.46h,t1/2(ka)=0.90h,T(peak)=3.24h,C(max)=433.9441mg/kg,AUC=3619.3877(mg/kg)·h,CL/F(s)=0.0954mg/kg/h/(mg/kg),V/F(c)=1.0274(mg/kg)/(mg/kg)。结果表明川芎挥发油ip及ig给药其体存量的表观药动学过程均符合一室开放模型,绝对生物利用度为6002%。
We assayed apparent pharmacokinetic parameter of Chuanxiong rhizome volatile oil with taking the ache effect as index and basing on the axiom of pharmacological effect method.The dosage is 52mg/kg(ip),596mg/kg(ig).The pharmacokinetic parameters (ip) are as follows:Ke=0.1138h 1 ,ka=3.3261h 1 ,t 1/2 (ke)=6.06h,t 1/2 (ka)=0.21h,T(peak)=1.05h,C(max)=50.6255mg/kg,AUC=526.2315(mg/mg)·h,CL/F(s)=0.1637mg/kg/h/(mg/kg),V/F(c)=0.9114(mg/kg)/(mg/kg),the pharmacokinetic parameters (ig) are as follows:ke=0.0929h 1 ,ka=0.7693h 1 ,t 1/2 (ke)=7.46h,t 1/2 (ka)=0.90h,T(peak)=3.24h,C(max)=433.9441mg/kg,AUC=3619.3877(mg/kg)·h,CL/F(s)=0.0954mg/kg/h/(mg/kg),V/F(c)=1.0274(mg/kg)/(mg/kg).The results showed that the apparent pharmacokinetic process of internal dose is consistent with one compartment model,Fabs=60.02%.
