摘要
腹腔注射内毒素所致大鼠急性内毒素血症时肝组织、肝细胞及肝枯否氏细胞内PAF含量显著升高,肝枯否氏细胞培养上清液中PAF含量也显著升高,但肝细胞培养上清液中PAF未见明显升高,并见肝组织MDA明显增加,ATP显著降低。银杏内酯B5mg/kg于攻毒前15分钟ip可显著提高肝组织ATP的含量,降低MDA的水平,但不影响PAF的升高。表明PAF可能是内毒素血症时肝功能损害的重要介质之一,枯否氏细胞可能是肝内PAF的主要来源,银杏内酯B可能是一个内毒素休克时有价值的侯选治疗药物。
Aimes: The experiment was designed tostudy the changes of intrahepatic PAF production, the reactions of different cells under stimulation by endotoxin and especially to observe the protective effects of PAF receptor antagonist Ginkgolide B during endotoxemia. Materials and methods: It was carried out by both in vivo and in vitro. In vivo:Rats were divided into contol group (n=5), endotoxemia group (n=30) and end otoxemia with Ginkgolide B pretreatment group (n=25).Rats were decapitated and sampled at phases of 1,3,5, 8 hours after drugs administration.Parameters being observed included hepatic P AF,MDA and ATP levels.In vitro: Rats'liver were in situ perfused at time phases as control and 1,3,5,8 hours after endotoxin administration. The separated and purified hepatocytes of Kupffer cells were cultured in order to determine the intracellular PAF levels or PAF levels in supernatum. Results: PAF levels of the liver tissues or of the intrahepatocytes and in the Kupffer cells increased remarkably during acute endotoxemia.At mean time, PAF contents of the supernatum of cultured Kupffer cells increased markedly as well. But PAF contents of the supernatum of cultured hepatocytes changed unremarkably. In addition, in the liver tissues MDA levels increased and ATP contents decreased obviously.Pretreat with Ginkg olide B could effectively reverse the intoxicative effects of the endotoxin,and increase ATP levels of the liver tissues and decrease MDA contents,but it did not affect the increasement of PAF levels. Conclusion: PAF might be one of the important mediators that could induce damages of hepatic functions. Kupffer cell may be the main resourses of the intrahepatic PAF.Well,PAF receptor antagonist Ginkgolide B is one of the important candidate for therapeutical treatment of endotoxic shock.
