骨肉瘤及其周围肌肉组织中NOS和NO的测定及其意义

摘要 目的 研究骨肉瘤及其周围肌肉组织中一氧化氮合酶 (NOS)活性与一氧化氮 (NO )含量之间的关系。方法 采用分光光度法对 37例骨肉瘤组织 ,分别测定其中NOS活性和NO含量 ,同时分别测定肿瘤周围正常肌肉组织中NOS活性和NO含量 ,然后进行相应的统计学分析。结果 肿瘤组织和肿瘤周围正常肌肉组织中NOS活性分别为 ( 0 .15± 0 .0 5 )U /mg蛋白质和( 1.88± 0 .2 7)U /mg蛋白质 ,两者比较有非常显著性差异 (P <0 .0 1) ,NO含量平均分别为 ( 46.43± 2 5 .98) μmol/L和 ( 49.0 8±1.5 0 ) μmol/L ,两者差异不明显 ;转移性瘤组织与非转移性瘤组织中NOS活性分别为 ( 0 .16± 0 .0 4)U /mg蛋白质和 ( 0 .13±0 .0 6)U /mg蛋白质 ,两者差异不明显 ,NO含量分别为 ( 36.44± 2 7.2 1) μmol/L和( 5 6.43± 2 0 .82 ) μmol/L ,两者比较有显著性差异 (P <0 .0 5 ) ;转移性瘤周围正常肌肉组织与非转移性瘤周围正常肌肉组织中的NOS活性分别为 ( 1.2 3± 0 .34 )U /mg蛋白质和 ( 2 .5 4±0 .5 5 )U /mg蛋白质 ,两者比较有显著性差异 (P <0 .0 5 ) ,NO含量分别为 ( 40 .72±1.5 4) μmol/L和 ( 5 7.46± 2 .18) μmol/L ,两者比较有显著性差异 (P <0 .0 5 )。 Objective To study the relations between the activities of nitric oxide synthase (NOS) and the nitric oxide (NO) contents in osteosarcoma and its surrounding normal muscle tissues.Methods 37 osteosarcoma specimens and their surrounding normal muscle tissues were gathered.The activities of NOS and the contents of NO were measured through the colorimetric method.Statistical analysis was performel by use of SPSS software.Results The average activities of NOS were(0.15±0.05) U/mg protein and(1.88±0.27) U/mg protein in osteosarcoma specimens and its surrounding normal muscle tissues resepectively.The average contents of NO were(46.43±25.98) μmol/L and(49.08±1.50) μmol/L in osteosarcoma specimens and its surrounding normal muscle tissues respectively.There was obviously difference in NOS activelies but not in NO contents.The activities of NOS in metastatic and nonmetastatic osteosarcoma were(0.16±0.04) U/mg protein and(0.13±0.06) U/mg protein respectively.The contents of NO in these two tissues were(36.44±27.21) μmol/L and(56.43±20.82) μmol/L.There was obviously difference in NO contents but not in NOS activetes.The activities of NOS in the surrounding normal muscle tissues of metastatic and nonmetastatic osteosarcoma were(1.23±0.34) U/mg protein and(2.54±0.55) U/mg protein.The contents of NO in these two tissues were( 40.72 ± 1.54) μmol/L and(57.46±2.18) μmol/L.There were obviously differences both in NOS and NO.Conclusion NO in osteosarcoma was generated by osteosarcoma itself and it's surrounding normal muscle tissues.Higher NOS activity would diminish the tendency of metastasis of osteosarcoma.Detection of NOS activity and NO content would have some clinic significance in therapy and prognosis of osteosarcoma.
