人结肠癌细胞裸鼠肝转移模型及其血中CK20 mRNA表达的研究 被引量:7

摘要 目的 :建立人结肠癌裸鼠肝转移模型 ,检测血中 CK2 0 m RNA表达 ,应用于结肠癌肝转移的防治研究。方法 :BAL B/ C裸鼠 12只 ,腹腔内注射戊巴比妥钠麻醉 ,经脾脏注入指数生长期的人结肠癌低分化腺癌细胞悬液( L S174 t) 0 .1m l,含细胞数 1× 10 6 个 ,切除脾脏 ,喂养 2 0天后 ,建立人结肠癌裸鼠肝转移模型。随后 ,心脏采血检测血中 CK2 0 m RNA表达 ,处死裸鼠观察腹腔内肿瘤生长和肝转移癌灶数目及大小 ,作病理组织学检查。采用巢式逆转录聚合酶链反应 ( RT- PCR)法 ,检测裸鼠血中 CK2 0 m RNA表达。结果 :12只裸鼠均建立人结肠癌裸鼠肝转移模型 ,无1只死亡。初期活跃如常体形无改变 ,10天后裸鼠出现消瘦、精神差 ,摄食量减少。裸鼠尸解肝脏表面均有灰白色转移癌结节形成 ,其它脏器未发现转移癌灶。病理组织学示肝转移癌灶具有人结肠癌低分化腺癌特征。血中 CK2 0 m RNA均阳性表达。结论 :经脾脏注入 L S174 t细胞悬液可建立人结肠癌裸鼠肝转移模型 ,裸鼠血中 CK2 0 m RNA阳性表达 ,证实血中存在着结肠癌细胞 ,是形成肝转移灶的条件。 Objective: To develop a human colon carcinoma liver metastases model for experimental studies in nude mice ,CK20 mRNA in blood of nude mice was detected.Methods:liver metastases were established in twelve BALB/c nude mice by intrasplenic injection of colonic adenocarcinoma cells(Ls174t) with 1×10 6 cells in 0.1ml solution,the spleen wasn't left, after twenty days every mouse was killed.Blood was collected ,The growth and the number of the liver metastases were evaluated, the liver metastases were studied histologically. Using nested reverse transcript polymerase chain reaction(RT-PCR) technique CK20 mRNA in blood of 12 nude mice with liver metastases from colon carcinoma was detected. Results:During the whole experiment,all mice inoculated had lived,after 10 days, they became worse and worse. All mice inoculated with LS174t cell lines developed liver metastases,other organic metastases weren't observed.the pathological results showed that tumour cells of liver metastases accorded with poorly differentiated colonic adenocarcinoma,the expression of CK20 mRNA in blood of them was 12/12. Conclusion:Intrasplenic injection of Ls174t cells is a reliable model for producing colonic cancer liver metastases and should prove valuable in experimental study. The expression of CK20 mRNA in blood is a foundation of liver metastases from colon carcinoma in nude mice.
