蛇床子总香豆素的测定及其提取条件的正交设计优选 被引量:10

摘要 目的 建立蛇床子总香豆素的RP -HPLC测定方法 ,探讨其提取分离的优化条件。方法 以蛇床子素、欧前胡素为定量指标成分 ,通过对蛇床子浸出、水煎、超声等提取方法的比较与筛选 ,确定乙醇浓度、水浴温度、超声时间、浸渍时间 ,并以L9(34 )正交设计表安排试验。结果 蛇床子药材细粉加 2 0倍量 75 %乙醇5 5℃水浴温度下 4 3Hz超声提取 90min为蛇床子总香豆素的最佳提取工艺。结论 优选的超声提取条件效率高 ,可增加蛇床子药材的利用率。 Objective: To establish the RP-HPLC method for the determination of total coumarins in Fructus Cnidii and to optimize its extraction conditions by orthogonal design. Methods: With osthol and imperatorin content as the indexes, ethanol concentration, water-bath temperature, ultrasound time and soaking time were screened and L 9 (3 4) orthogonal test was applied. Results: The optimal extraction conditions were as follow: ultrasound extraction in 55℃ water-bath at 43 Hz frequency with 20-fold 75% ethanol for 90 minutes. Conclusion: The optimized ultrasound condition is the best for the extraction of coumarins and increase the availability of Fructus Cnidii
