脑创伤与缺血后大鼠脑内NOS阳性细胞变化和神经元损害的形态学观察 被引量:1

摘要 本实验应用NADPH—d组织化学技术结合尼氏染色观察了大鼠皮层针刺损伤及全脑缺血/再灌注术后皮层、海马及下丘脑室旁核等敏感区域神经元受损与NOS阳性细胞的变化。结果如下:(l)缺血组:术后1-3天,见散在的神经细胞固缩坏死,轮廓漠糊;皮层及海马CAl、CA。区NOS阳性细胞增多,尤以海马CA。区、下丘脑室旁核及周边区域明显;阳性细胞胞体多小而圆、突起较短、胞质浅染、同时也见少数胞体大,染色深,突起长的阳性细胞。术后5-14天,神经元数目减少,尤以海马显著;皮层及海马CAl、CAs区的NOS阳性细胞仍较正常多,细胞体积大,着色深,突起长;而下丘脑室旁核周边区域的NOS阳性细胞几乎完全消失。(2)损伤组:术后l-3天,伤道周围见大量红细胞渗出及死亡细胞碎片,NOS阳性细胞数目及形态较正常对照无显著变化;术后5-7天,伤道周围仍见坏死及固缩的神经细胞碎片,在颞皮质区、海马CA。区与下丘脑室套核区可见NOS阳性细胞数目明显增多,胞体染色加深,并见长的突起;术后14天,伤道周围神经元稀疏,但上述区域的NOS阳性细胞数目求见减少。本实验证实不同损伤时NOS阳性细胞的反应时相存在差异,在脑缺血1一3天内即达到高峰并维持到2周左右,而针刺损伤则发生于5—7天;并提示NOS阳性细胞与神经元?
