Previous studies have shown that the typical nucleus can be assembled around the naked lambda DNA in cell free Xenopus egg extracts. However, it is not very clear if the nucleus may be formed around the other exogenous naked DNA in the cell-free system. In our experiment, we used the purified adenovirus DNA to reconstitute the nucleus in the nuclear reconstitution extracts and found that typical nuclei can also be assembled around the added DNA. Generally, 2.500 new nuclei are formed in one egg's extract. The most assembled nuclei are 10-15 μm in diameter, and possess a double-membranized nuclear envelope with typical nuclear pore complex structures and chromatin-like materials. In control samples of no added DNA, no nucleus is formed. So, we get a conclusion that the purified adenovirus DNA can also be used to induce the nuclear reconstitution in cell-free extracts of Xenopus eggs.
