LAK细胞及活化胚胎胸腺细胞对脑胶质瘤细胞的体外作用

摘要 目的 探讨体外培养后人胚胎胸腺细胞 (ETC)表面抗原的转变及进行移植的时机 ,比较不同源LAK细胞、活化ETC联合与TJ90 5细胞的体外作用效果。方法 对不同处理的人胚胎胸腺悬液培养前后观察 ,测定细胞表面抗原 ;进行LAK细胞、活化ETC体外杀瘤活性比较。结果 应用特殊培养液培养 ,可以使人胚胎胸腺上皮细胞生存2 8天 ,分离后悬液比混悬液细胞表面抗原早期达稳定 (CD4/CD8=2 .2 6 ) ;胸腺LAK细胞对TJ90 5细胞的杀伤作用与脾LAK细胞无显著性差异。结论 活化ETC在体外对肿瘤细胞无抑制作用 ,不同源LAK细胞和活化ETC联合对TJ90 Objective To probe into the changes of the surface antigens of embryo thymocytes(ETC) after cultured in vitro and the transplantation opportunity of thymocytes,to compare the affect of TJ905 glioma and LAK cells of different source combined with activated ETC in vitro .Methods By observing the cell growth pattern and survival of human ETC before and after different handled culture in vitro , we could determine the changes of cell surface antigens and compare the killing glioma activity of LAK cells and activated ETC. Results Embryo thymus epithelial cell of human cultured by applying improved methods could live 28 days. The surface antigens of the severed suspension came to stabilization earlier than those of the mixed suspension (CD4/CD8=2.26). The killing TJ905 glioma activity of spleen LAK, compared with thymus LAK, had no obvious difference.Conclusion Activated ETC have inhibition to glioma cells in vitro . LAK of different source combined activated ETC have killing TJ905 glioma activity.
