抗CD_3单链抗体的构建及核酸序列分析

摘要 目的探讨用抗CD3抗体重链可变区与轻链可变区基因,经重叠延伸拼接连接形成单链抗体。方法用DNA序列分析及动态模拟构象分析,并与相关软件分析,推导Scfv基因编码的氨基酸序列。结果该基因全长732bp,三维构象稳定,兼容性分数S>0。将单链抗体基因克隆于载体中,在强启动子作用下,目的蛋白的表达量约占菌体蛋白总量的20%。结论重链可变区与轻链可变区经短肽连接而成的单链抗体,具有分子量小,免疫原性低的特性,具有较广的应用前景。 Objective To investigate single chain antibody linked by the variable regions genes of heavy chain(VH)and light chain of anti-CD 3 antibody by means of splicing,overlapping and exten sion(SOE).Methods The DNA sequence of the anti-CD 3 single chain Fv was analysed by DNA se quence analysis and dynamic simulation conformation with relative software,derivating the amino-acid se quences coded by ScFv gene.Results The anti-CD 3 single chain Fv(ScFv)gene has732bp,3D-conformation stabi lization,com patibility S>0.The expression level of aim protein cod ed by the gene is about 20%of total bacte ria pro tein level.Con clusion anti-CD 3 ScFv-coding protein has small molecule weight ,low im munogenic ity,thus has wider prospect of application.
