恶性疟原虫DNA疫苗在小鼠组织中的表达

摘要 目的 探讨恶性疟原虫 FCC- 1/ HN株环子孢子蛋白 (CSP) DNA疫苗在小鼠组织中的表达。 方法 用盐酸布比卡因对 BAL B/ c小鼠的骨骼肌进行了预处理 ,然后在同一部位注射重组真核表达质粒 p BK/ CSP。分别在免疫后 4周和 8周应用间接免疫酶法检测免疫小鼠注射部位肌肉组织中重组蛋白 CSP的表达。 结果 小鼠经 DNA疫苗免疫 4周和 8周后肌肉组织及其周围结缔组织均呈现特异性阳性反应。 结论 疟疾 DNA疫苗免疫小鼠后 ,肌肉组织及其周围结缔组织有重组 CSP蛋白的表达。 Objective To explore the recombinant protein expression status in muscle tissue of mice vaccinated by DNA vaccine of circumsporozoite protein (CSP) of Plasmodium falciparum FCC 1/HN strain. Methods The recombinant pBK/CSP was injected into BALB/c mice by intramuscular route, with pretreatment of the muscle tissue of the injected sites by bupivacaine hydrochloride, and the expression product of DNA vaccine was detected with indirect immunohistochemistry assay after 4 weeks and 8 weeks immunization. Results The specific positive reaction of the indirect immunohistochemistry assay were observed in muscle tissue and its adjacent connective tissue in immune group.(For Fig. 1-4, to see back cover) Conclusion After vaccinating BALB/c mice with malaria DNA vaccine by injection muscle directly, it is obvious that the recombinant protein CSP could be expressed in the muscle tissue and connective tissue.
