摘要
目的 :观察抗血管内皮生长因子抗体对实验性肝癌的生长和肿瘤内部微血管生成的作用。方法 :在已建立的肝癌裸鼠皮下种植瘤的瘤灶旁 ,多次注射抗血管内皮生长因子抗体 ,用 CD3 1 的免疫组化标记肿瘤内部的微血管内皮细胞 ,用肿瘤的长轴×宽轴估计肿瘤的大小。结果 :在注射方案结束 1,2周后 ,同对照组相比 ,实验组的肿瘤大小受到明显抑制 (P <0 .0 0 1和 P <0 .0 5 ) ,2周后肿瘤内部的微血管密度明显降低 (P <0 .0 0 1)。结论 :抗血管内皮生长因子抗体能通过抑制肿瘤内部微血管的生成 。
Objective:To observe the effect of antibody against vascular endothelial growth factor (VEGF) in the growth of the human experimental hepatocellular carcinoma(HCC)and antiangiogenesis to tumour tissue Methods:To inject anti VEGF antibody s c at intervals,nearby the founded subcutaneous exnograft focals of the human experimental HCC in nude mice To mark the intratumoural endothelial cells of microvessels with immunohistochemical staining of CD 31 To estimate the tumour's sizes with the tumour's length multiplying the tumour's width Results:In one and two weeks later after completing all the injecting procedure,the tumour's sizes of the test group are significantly inhibited (P<0 001 and P<0 05,respectively)by comparison with the control group Two weeks after finishing injection procedure,the intratumoural microressel density of the test group is extremely decreased (P<0 001)by comparison with the control group Conclusion:The anti VEGF antiboby can suppress the growth of the human experimental HCC through inhibiting the intratumoural angiogenesis
