家兔纹状缘囊泡的制备及其对5-FU摄取的初步考察

摘要 目的 :研究药物在小肠亚细胞结构中的摄取机制。方法 :通过钙离子投入法制备家兔小肠纹状缘囊泡系统 ,透射电镜观察纹状缘囊泡的微观形态 ,运用福林酚法对其进行定量分析 ,通过快速渗滤法初步考察 5 -FU在纹状缘囊泡系统中的摄取。结果和结论 :制备得到的囊泡均匀圆整 ,粒径 0 .5 5± 0 .11μm,5 -FU在纹状缘囊泡的摄取过程包含主动转运与至少一个顺浓度梯度的被动过程。该囊泡适于研究药物在小肠亚细胞结构中的摄取。 AIM The purpose is to study the absorption mechanisms in the sub-cell structure of small intestine. METHODS Brush border membrane vesicles (BBMVs) was prepared from rabbits by the Ca 2+ precipitation procedure. The appearance of vesicles was observed by transmission electron microscopy. The contents of protein were measured by Folin method. The uptake of 5-FU by BBMVs was determined by the fast filtration techniques. RESULTS AND CONCLUSION The vesicles were spherical particles with size of 0.55±0.11μm. The uptake of 5-FU contained active transport and at least one concentration gradient-dependent passive process. The BBMVs model is suitable for the study of absorption mechanisms in the sub-cell structure of small intestine.
