PDTC对创伤性肺损伤大鼠肺组织TNFα、IL-8含量及其mRNA表达的影响 被引量:5

摘要 目的 观察PDTC对创伤后肺组织TNFα、IL-8含量及其mRNA表达的影响。方法80只大鼠分为创伤组(T组)、PDTC预处理组(P组)及对照组(C组),利用多功能小型生物撞击机致伤,观测伤后各组0.5、1、2、4、8 h肺组织TNFα、IL-8及其mRNA变化。结果 致伤后TNFα、IL-8含量及其mRNA表达明显升高,TNFα在伤后1h达到峰值,IL-8在伤后4h达到峰值;PDTC组 TNFα、IL-8含量及其mRNA表达显著低于创伤组(P<0.01)。结论PDTC可以通过抑制TNFα、IL-8 mRNA表达,减少致炎细胞因子的释放,减轻创伤后全身及肺组织的炎症损害。 Objective To investgate the effect of pyrrolidine dithiocarbamate (PDTC) on TNFα, IL-8 content and their mRNA expression in traumatic acute lung injury in rats. Methods A total of 80 Wistar rats were divided into traumatic group (T), PDTC pretreatment group (P) and control groups. The traumatic acute lung injury was obtained by multi-funtion minitype bio-impact machine with driving pressure of 350 kPa on the right superior lateral chest wall of rats. The contents of TNFα, IL-8 and the expression of TNFα, IL-8 mRNA were determined by ELISA or RT-PCR respectively. Results The TNFα, IL-8 content and the expression of TNFα, IL-8 mRNA in traumatic group were markedly increased, the peak of TNDα, IL-8 were at 1 h, 4 h after trauma respectively. The TNFα, IL-8 contents and the expression of TNFα, IL-8 mRNA were markedly lowerr in PDTC group(P < 0.01) than that in group T. Conclusion PDTC can alleviate the traumatic acute lung injury by inhibiting the expression of TNFα and IL-8 mRNA.
