大鼠肝脏缺血再灌注合并肝部分切除诱导肝再生模型中EGFR及CyclinD_1的表达 被引量:2

Expressions of EGFR and CyclinD_1 in rat model of liver ischemia/reperfusion and liver regeneration after partial hepatectomy

摘要 目的 观察大鼠肝脏缺血再灌注合并肝再生诱导模型中EGFR及CyclinD1的表达情况。方法 采用预先保留门静脉转流肝叶(约占全肝70%),阻断部分肝叶(约占全肝30%)血供,到预定观察终点后重新开放血供灌注缺血肝叶,同时切除门静脉转流肝叶(70%肝切除)模型,通过RT-PCR和免疫组化法分析EGFR及CyclinD1在大鼠肝脏缺血再灌注对肝细胞再生功能影响过程中的变化。结果 大鼠术后 12、24 h CyclinD1 mRNA及此后相应的蛋白表达水平随着缺血时间的延长而显著降低。术后 12 h内 EGFR蛋白表达水平随着缺血时间的延长而显著降低。结论 术后EGFR以及CyclinD1表达降低可能是肝脏缺血再灌注影响大鼠肝再生的原因之一。 Objective To observe the expression of EGFR and CyclinD1 in the model of liver ischemia/reperfusion with induction of liver regeneration by partial hepatectomy in rats. Methods Healthy male Wistar rats were subjected to segmental liver ischemia for 30, 60, 90 min respectively, ischemia liver lobes were then reperfused and non-ischemia liver lobes were resected (70% partial hepatectomy). Partial hepatectomy without prior liver ischemia was performed in the control group. By detecting expression of CyclinD1 and EGFR during corresponding days after opera- tion, we can evaluate the effect on liver regeneration after ischemia and reperfusion-injury in rat. Results With increasing in the duration of isch- emia and reperfusion the liver expression of CyclinD1 and EGFR became lower. Significant differences were observed between groups of N-PH and 60' PH, N-PH and 90' PH. Conclusion The low expression of EGFR and CyclinD1 may be one of the reasons that hepatic ischemia repertusion af- fects the hepatic regeneration of rats.
