D-丙~6-去甘^(10)GnRH乙基酰胺(GnRH-A)150μg/天能使正常大鼠卵巢GnRH受体含量增加;去垂体大鼠GnRH-A 15μg/天能使卵巢GnRH受体量增加,加用孕马血清75IU/天后,卵巢GnRH受体又显著减少,血中雌二醇、孕酮增加;在妊娠15天大鼠,GnRH-A 15μg/天同样引起卵巢GnRH受体增加,而血中雌二醇、孕酮则减少,胚胎死亡。GnRH-A对卵巢GnRH受体亲和常数没有影响。本实验结果提示,大剂量GnRH-A对大鼠卵巢GnRH受体有自身诱导作用,促性腺激素使卵巢GnRH受体降调节,GnRH-A终止15天龄大鼠妊娠的作用与卵巢GnRH受体含量增加有关,卵巢GnRH受体含量与血中雌二醇、孕酮水平呈负相关。
The ovarian GnRH binding capacity after treatment with D-Ala^6-des-Gly^(10)-GnRHethylamide(GnRH-A)150μg/day for 3 days in the intact adult rats was increased to81.97 fmol/mg protein(Saline control 46.02 fmol/mg protein).In the hypophysecto-mized—rats,the injections of GnRH-A 15 μg/day for 3 days caused an increase in theovarian GnRH binding capacity.In the H rats treated with GnRH-A 15μg/day,theinjections of PMSG 75 IU/day for 3 days brought about a decrease in the number ofovarian GnRH receptors while the levels of E_2 and P were markedly increased.Theovarian GnRH binding capacity was found to increase in the 15-day-pregnant ratsinjected with GnRH-A 15μg/day for 3 days.The rats had levels of serum E_2 and Pand death rate of fetus decreased. This results suggest that a high dose of GnRH-A can give rise to a self-induce-ment of its receptors in the ovary of the rat.The number of ovarian GnRH receptorsin the rat unper the effects of pituitary gonado tropinshow a down-regulation.The ter-mination of pregnancy of 15 pregnant rats treated with GnRH-A 15μg/day for 3 dayswas related to the increase in capacity of the ovarian GnRH receptor in rat.The capacityof the ovarian GnRH receptor and serum levels of E_2 and P in each treated rat isnegatively correlated(P<0.05).
