摘要
抗人精子单克隆抗体YWKⅠ和YWK-Ⅱ及抗兔精子单克隆抗体rSMP-B均能显著抑制人精子穿入去透明带金黄地鼠卵。在本文的小鼠体外受精实验中,获能精子先与10倍稀释的抗精子单克隆抗体作用10min后再与卵子混合。结果表明:三种抗精子单克隆抗体对小鼠体外受精均有明显的抑制,其中YWM-Ⅰ使受精率下降至16.9%。此外还发现YWK-Ⅱ及rSMP-B主要抑制小鼠精子穿入透明带,而YWK-Ⅰ除此之外还抑制穿入透明带后的精子进一步完成受精。
The inhibition assay of mouse in vitro fertilization(IVF)was used in this ar-ticle to study the effect of three anti-sperm monoclonal antibodies(MAbs)(i.e:anti-human sperm MAbs YWK-I,YWK-II and anti-rabbit sperm Mab rSMP-B)on fertil-ization.For this purpose,the capacitated mouse sperm was mixed with 10 times dil-uted MAbs for 10 minutes before mixing with eggs,Markedly lower penetration rateand fertilization rate were obtained than With the of contol group(SP 2/0 ascitesfluid), and MAb YWK-I had the strongest effect among these MAbs(fertilization ratedropped to 16.9%).Moreover,YWK-II and rSMP-B were found to inhibit mainlysperm penetration through the egg's zona,while YWK-I inhibited botlh the penetra-tion through the zona and the following fertilization process of the penetated sperm,
