摘要
目的 :研究大黄对烫伤和内毒素“二次打击”模型大鼠肝细胞线粒体功能的影响。方法 :采用大鼠背部烫伤模型 ,烫伤后 12小时静脉内给予内毒素 2 0 m g/ kg进行“二次打击”。分离肝细胞线粒体 ,监测肝细胞线粒体内 ATP水平、细胞色素 C含量和能荷。结果 :烫伤能引起肝细胞内 ATP含量明显减少 ,细胞色素 C和能荷明显下降 ;内毒素打击后上述指标降低更显著 ,且此过程随时间而加重 ,各时间点与正常对照组均有显著性差异 (P均 <0 .0 0 1) ;大黄可明显提高脓毒症大鼠细胞内 ATP含量和能荷值 ,减少肝细胞内细胞色素 C的丢失 ,与单纯烫伤组和脓毒症组比较有非常显著性差异 (P均 <0 .0 0 1)。结论 :烫伤和内毒素“二次打击”能导致肝细胞内 ATP和细胞色素 C含量明显减少 ,能荷下降 ,而大黄能明显提高肝细胞内 ATP和细胞色素 C含量和能荷 ,改善肝细胞线粒体的呼吸功能。
Objective:To investigate the effects of rhubarb on the function of mitochondria of hepatic cell in posttrauma sepsis.Methods:One hundred and eight SD rats with 25% Ⅲdegree burns on back hit by endotoxin at a dose of 20 mg/kg 12 hours after the burn injury.Mitochondria was isolated from the hepatic cells.The level of ATP,cytochrome C,and energy charge in hepatic cells were applied to evaluate the function of mitochondria.Results:The level of ATP,cytochrome C,and energy charge in hepatic cells was decreased after burn injury,and deteriorated further after the second hit by endotoxin on a time manner.There were significant differences compared to the controls (all P <0 001).However,rhubarb could obviously increase the level of ATP,cytochrome C and energy charge in hepatic mitochondria,the differences were significant compared to the burn and septic group (all P <0 001).Conclusions:Burn injury and the postburn sepsis,hit by endotoxin,can damage the function of mitochondria in hepatic cells.Whereas,rhubarb obviously improved the mitochondrial respiration of hepatic cells in the postburn sepsis.
