摘要
本文对一株人抗人A-血型物质单克隆抗体,用定量免疫沉淀法以及ELISA研究其与多种单糖、双糖及寡糖的反应性,从而确定了其结合部位的结构特异性。实验发现其结合部位互补于含有双分子岩藻糖残基的A-t糖:这一研究进一步强调了含有双分子岩藻糖残基的A血型抗原决定簇的重要性。
This is the first human monoclnal antibody against A blood group substance, generated by Epstein-Barr virus trans-formation of huma lymphocyte isolated from splenic tissue after activation in vitro with group A blood cells.the antibody has been characterized by quantitative precipitin assays and the fins structure of its combining site has been mapped by quantitative ELISA assays by using A-active oligosaccharides to inhibit the binding of anti-A and A blood group substance. The human monoclonal anti-A strongly precipitated various A-active blood group substances, but not B, H, Lea, and the precusor of Ii substances.The combining site was most complemntary to the difucose containing A-Hep tasaccharide:GalNAcα1→3Galβ1→3GlcNAcβ1→3Galβ1→4Glc Fuca FucaThe se studies futher emphasize the importance of difucosyl determinants.
