摘要
外源基因在原核细胞中的分泌表达仍然是很活跃的研究领域。Fujimoto等系统地分析比较了大肠杆菌内毒素ST-Ⅰ、ST-Ⅱ、LT-A、LT-B信号肽在人表皮生长因子(hEGF)分泌表达中的作用指出,由ST-Ⅱ信号肽组成的表达载体在大肠杆菌中能精确加工并使hEGF这样小的多肽成功地分泌到细胞周质及培养基中。
The signal peptide gene of heat-stable enterotoxin Ⅱ of E. coli waschemically synthesized according to codon usage in E.coli. A signal pe-ptide gene cartridge plasmid pST-Ⅱ was constructed by inserting the si-gnal peptide gene into BamH Ⅰ/PvuⅡ sites of pBR322.The gene block caneasily be excised and transferred to other genetic systems for construc-tion of secretion expression vector by digestion with BamH Ⅰand XhoⅠ/Kpn Ⅰ. The GCGC sequence at 3′-end of ST-Ⅱ signal peptide gene pro-vided Hha Ⅰ/HinpⅠcut sites which should be convenient for insertingtarget gene in frame.
