摘要
目的 探讨武汉地区肾综合征出血热重型患者感染的汉坦病毒M片段G1基因变异的情况。方法 用RT PCR对 34份肾综合征出血热患者发病 1周内血清中的汉坦病毒G1基因扩增 ,并选取 7份重型 ,1份轻型及 1份中型患者的PCR产物测序 ,分别与标准株HV114和R2 2株相应序列比较。结果 1.Ⅰ型重型患者G1区在 15 1~ 2 10位核苷酸有意义突变的频率很高 ,而轻型患者则无此改变。 2 .Ⅱ型重型患者G1区 5 49位 ,5 5 2位和 6 92位核苷酸变异引起编码氨基酸的改变相同 ,中型患者无此特点。结论 这些核苷酸变异可能与病毒毒力增强有关 ,从而引起临床重症表现。
Objective To study the G1 gene variation in severe type patients with hemorrhagic fever with renal syndrome. Methods Thirty four serum samples of clinically diagnosed patients with hemorrhagic fever with renal syndrome were amplified by typing RT-PCR, then the PCR products of 7 severe type cases, 1 moderate type case, and 1 mild type case were sequenced and compared with the corresponding sequences of HV114 and R22 respectively to find whether there were hot variations in severe type cases in Wuhan district. Results There were high frequent sense mutations between 151~210bp on G1 gene in severe case, but not in mild case of type Ⅰ. In severe cases of type Ⅱ, the different nucleotide mutations on 552, 549, and 692 resulted in an identical code of amino acid. Conclusion The sense mutations between the 151 and 210 bp of G1 gene in the cases of type Ⅰ, and on sites of the 549, 552 or 692 bp in the cases of type Ⅱ may result in the enhancement of virulence.
