摘要
玉米授粉后12天的幼胚,在MS附加2,4—D2mg/1和12%蔗糖培养基上,可诱导产生高频率的盾片愈伤组织。以1—4—氧赖氨酸(OL)、S—2—氨乙基—1—半胱氨酸(AEC)和赖氨酸+苏氨酸(LT)为选择剂,适宜选择浓度分别为150mg/1、0.5mM和0.7mM。同工酶与玉米籽粒赖氨酸含量比较分析结果表明,未开花雄穗酯酸同工酶具有高赖氨酸表型的特征酶带,可作为突变体鉴定的简便方法。
The young embryo from corn 12 days after pollination could be induced to form callus at high frequency on the MS medium with 2,4--D 2mg/l plus 12% sugar.When 1--4--oxalysine (OL),S--2--aminoethgl--1--cysleine(AEC) or lysine plus threonine(LT)were used as selection agents with 150mg/l, 0.5mM and 0.7mM as the suitable concentration respectively. Comparative analysis of lysine content of corn seed and electrophoretogram of isozymes showed that the isozymes of esterase from tassel before blooming had characteristic belt of high lysine, which could be used as a simple method for identifying mutants.
