摘要
将正常血小板与用胶原刺激的血小板的胞浆部分与膜部分进行SDS-聚丙酰胺凝胶电泳(PAGE),并测定各部分的肌动蛋白激活的肌球蛋白Mg~(2+)-ATPase活力(AM-Mg~(2+)-ATPase)后发现:正常血小板膜部分所连接的肌球蛋白与肌动蛋白分别占血小板中此两种蛋白总量的26.40%)和31.55%。血小板受胶原刺激后则分别变为87.83%和42.17%。胞浆中的AM-Mg~(2+)-ATPase活力从正常的1.43±1.37nmolPi/min(每毫克肌球蛋白)升高到刺激后的4.66±1.51nmol Pi/min(每毫克肌球蛋白)。这些结果表明;血小板的胶原受体被结合后,有大量肌球蛋白和较少量肌动蛋白连接到胞膜上。另一方面,胞浆中的AM-Mg~(2+)-ATPase活力也明显增加。这些变化可能与血小板的形态变化和功能变化有密切关系。
SDS-polyacrylamide gel electrophoresis and actin activated myosin Mg2+-ATPase (AM-Mg2+-ATPase) measurements were performed to determine the distribution of cytoskeleton components in the membrane and oytosol frac bions of resting and activated platelets.Results showed that the content of myosin and aotin in the membrane fraction of resting platelets was 26.4% and 31.55% respectively.Upon activation with collagn,they were increased to 87.33% and 42.17% respectively.AM-Mg2+-ATPass activity of the oytosol fraction was 1.43nmol Pi/min (mg myosin) in resting platelets,and 4.66 nmol Pi/min (mg myosin) in activated platelets,From these results it may be seen that when the platelets were stimulated with collagen,the percentage of membrane attached aotin and myosin and oytosol AM-Mg2+-ATPase activity increased significantly,indicating the redistribution of aotin and myosin between membrane and cytosol fractions and AM-Mg2+-ATPase were involved in platelet function alterlation in collagen stimulation.
