白细胞介素18重组腺病毒对小鼠腹腔巨噬细胞免疫功能的影响 被引量:6

摘要 目的 :研究白细胞介素 1 8重组腺病毒 ( Adms IL -1 8)对小鼠腹腔巨噬细胞免疫功能的影响 ,并对其机理作一探讨。方法 :重组腺病毒腹腔注射 BAL B/ C小鼠 ,每周注射 1次 ,共 2次。 2周后制备腹腔巨噬细胞 ,经 L PS刺激 ,采用 EL ISA法检测培养上清 m IL-1 8、m IL-1 β、m TNFα的水平 ;酶还原法检测 NO的含量 ;L DH释放法检测巨噬细胞的杀伤活性。结果 :Adms IL -1 8注射后 ,小鼠腹腔巨噬细胞高效表达 m IL-1 8,同时巨噬细胞分泌 m IL-1 β、m TNFα、NO水平明显增加 ,并增强巨噬细胞的杀伤活性 ,与其它对照组比较均 P<0 .0 1。结论 :小鼠腹腔注射 Adms IL-1 8,能显著增加巨噬细胞表达 m IL -1 8、m IL -1β、m TNFα、NO,增强巨噬细胞的杀伤活性。 Objective: To study the effect of recombinant Adenovirus mediated Interleukin 18 gene (AdmsIL 18) transfection on immune response of peritoneal macrophages in mice and to explore its machanism. Methods: BALB/c mice were injected with AdmsIL 18 twice weekly. After 2 weeks, the peritoneal macrophages were isolated, cultured in vitro and stimulated by LPS. Then mIL 18, mIL 1β and mTNFα in culture supernatants were detected with ELISA and the level of NO was measured with enzyme reduction method. Meanwhile cytotoxicity of macrophages was assayed by LDH release method. Results: mIL 18 is expressed in mouse macrophages after injection of AdmsIL 18. The levels of mIL 1β, mTNFα and NO in macrophages were higher than those in controls ( P <0.01). Cytotoxicity of macrophages was also enhanced remarkably. Conclusion: AdmsIL 18 can significantly increase the levels of mIL 18 , mIL 1β, mTNFα and NO in peritoneal macrophages of mice as well as enhance the cytotoxicity.
