雌/孕激素对围着床期小鼠子宫内膜明胶酶表达的影响 被引量:3

摘要 目的 :了解雌、孕激素对围着床期小鼠子宫内膜明胶酶表达的影响。方法 :应用酶谱法检测经雌 /孕激素处理的早孕期及去卵巢后小鼠子宫内膜明胶酶的表达。结果 :明胶酶-B的表达总量在早孕 d 1~ 4明显下降 ,但 d 5 (植入开始 )骤然上升 ,同时活性型明胶酶 -A也相对增多。去卵巢后明胶酶表达明显减弱 ,且未能测出明胶酶 -A活性 ;明胶酶 B表达在雌激素组增强 ,孕激素组及雌 /孕激素处理组均减弱 ,经雌 /孕激素处理后仍未测出明胶酶 -A的活性。结论 :明胶酶的表达受雌激素上调、孕激素下调 。 Objective: To investigate the regulation of estradiol(E 2) and progesterone(P 4) on gelatinases in periimplantation. Method: The expression of gelatinases in the endometrium of mice during early pregnancy and treated by ovary hormones after ovariectomy were detected by zymography. Results: The expression of Gelatinase B(MMP9) decreased significantly through days 1~4, but on day 5 it highly increased in early pregnancy. The total of Gelatinase A(MMP2) had no significant change through days 1~5, but the active Gelatinase A increased relatively on day 5. In the ovariectomy group, only Gelatinase B expression can be detected and its expression was induced by E 2, but reduced by P 4 and E 2+P 4; There was no Gelatinase A bands in control group, but after treated by E 2, P 4 and E 2+P 4, Gelatinase A bands still can not be detected. Conclusion: The expression of Gelatinase B is up regulated by estradiol and down regulated by progesterone. Gelatinase B may attribute to the endometrium window for implantation and play an important role when embryo penetrates the basement and ECM of endometrium in implantation. Active Gelatinase A may also participate in the degrading and remodeling of ECM during the implantation.
