摘要
目的 :探讨 c FL IP(细胞型 Fas相关死亡区域蛋白样白介素 - 1β转换酶抑制蛋白 ) m RNA在大肠癌细胞中的表达及其与 Fas抗原 (CD95 )表达的相关性。方法 :应用 RT- PCR方法检测 3株人大肠癌细胞株中 c- FL IP m RNA的表达 ,采用间接免疫荧光标记流式细胞术检测大肠癌细胞 Fas抗原表达。结果 :SW1116及 SW6 2 0细胞株 c- FL IP m RNA表达阳性并相对高水平表达 Fas抗原 ,而 L o Vo细胞株 c- FL IP m RNA表达阴性并低水平表达 Fas抗原。结论 :在大肠癌细胞中 ,c- FL IP m RNA的表达趋势与 Fas抗原一致 ,提示 c- FL IP可能在抑制 Fas抗原诱导的细胞凋亡中发挥重要作用。
Objective:To investigate the expression of c-FLIP mRNA in colorectal cancer cells and its relationship with Fas antigen(CD95)expression.Methods:RT-PCR was employed to detect the expression of c-FLIP mRNA and indirect immunofluoresence label flow cytometry was utilized to assay the Fas antigen expression in 3 colorectal cancer cell lines.Results: c-FLIP mRNA was positive in SW1116 and SW620 cell lines and negative in LoVo cell lines.The expression of Fas antigen was much higher in SW1116 and SW620 cell lines than that in LoVo cell lines.Conclusion:The expression of c-FLIP mRNA had positive correlation with the expression of Fas antigen in colorectal cancer cells,and c-FLIP may significantly inhibit the apoptosis induced by Fas in colorectal cancer cells.
