摘要
目的 探讨宁得朗 (NIDRAN)对人脑胶质瘤细胞凋亡的诱导作用 .方法 用含 10 μmol· L- 1 NIDRAN的 RPMI16 40培养液 ,作用于 SHG44细胞 3h,收集细胞用于形态学检测 ,DNA电泳及流式细胞分析 .结果 1形态学观察 :大部分瘤细胞体积缩小 ,核固缩 ,染色质浓缩在核模边缘呈月牙状聚集 ,有凋亡小体形成 . 2 DNA电泳 :在 80 0 bp以下可见明显的 DNA梯状带 . 3流式细胞分析 :出现典型的细胞凋亡峰 ,凋亡细胞占细胞总数的 32 .1% .结论 NIDRAN可诱导人胶质瘤细胞发生凋亡 .
AIM To investigate the effect of Nimustine hydrochloride (NIDRAN) to induce the apoptosis of human cultured glioma cells. METHODS SHG44 cells, human glioma cell line, were treated with RPMI 1640 culture fluid containing 10 μmol·L -1 NIDRAN for 3 h. Then these cells were observed morphologically, by DNA electrophoresis and flow cytometry analysis respectively. RESULTS ① The cells became smaller, with nucleus of cells shrinking, chromatinaggregating at the margin of nuclear membrane and apoptotic body formed. ② DNA ladder under 800 bp was found by electorphoresis. ③ The typical apoptotic peak was observed by flow cytomery analysis and apoptotic cells accounted for 32.1%. CONCLUSION NIDRAN can induce the apoptosis of human cultured glioma cells.
