利用多聚赖氨酸和鲎试剂检测细菌内毒素

STUDY ON DETECTION OF ENDOTOXIN USING POLY-L-LYSINE AND LIMULUS AMEBOCYTE LYSATE

摘要 目的 :建立内毒素定量检测方法。方法 :利用多聚赖氨酸交联琼脂糖将内毒素从含干扰物的样品中分离出来 ,再以基质偶氮显色鲎试剂定量检测被吸附分离出的内毒素。结果 :多聚赖氨酸交联琼脂糖能有效地吸附样品中的内毒素 ,但不减弱内毒素的鲎试剂反应性。本法能较好地克服 β 1,3 D glucan、氨基酸、抗生素及血浆蛋白等对鲎试验 (LAL)的干扰 ,并具有较高的准确性和重复性。结论 :该法是一种简便、特异的内毒素定量检测方法 。 OBJECTIVE:To establish a simple,specific quantitative assay determination of endotoxins.METHODS:Endotoxins were quantitatively absorbed on poly L lysine cross linked agarose from all kinds of solutions containing LAL inhibiting or enhancing substances and the absorbed endotoxins reacted with the LAL chromogenic reagents.RESULTS:Poly L lysine cross linked agarose can effectively absorb endotoxins from all kinds of samples and the absorbed endotoxins show enough activity for LAL. This assay can eliminate the effect of interference substance such as β 1,3 D glucan,amino acid,antibiotics and plasm protein on LAL,and has high reproducibility and accuracy.CONCLUSION:This assay is a simple and specific quantitative assay for determination of endotoxins can be utilized for detecting endotoxin in a solution containing LAL inhibiting or enhancing substance such as amino acid and antibiotics instead of requiring employment of the more common LAL technique
