摘要
目的 :探讨高聚生、干扰素 α 2b对不同膀胱癌细胞株的生长以及对HLA DR及HLA ABC表达的影响。方法 :以不同膀胱癌细胞株为研究对象 ,采用细胞培养及MTT比色法 ,检测时间、浓度量效关系 ;以ID50 时的药物浓度作用于不同膀胱癌细胞株 72h后 ,流式细胞仪检测高聚生、干扰素 α 2b对膀胱癌细胞株HLA DR及HLA ABC表达的影响。结果 :高聚生对膀胱癌细胞的生长表现为促进作用 ,并呈量效关系 ,干扰素 α 2b表现为抑制肿瘤细胞生长 ,随着时间延长 ,抑制作用更为显著 ;所测 3个细胞株均无HLA DR抗原表达 ,高聚生、干扰素 α 2b对HLA DR的表达均无影响 ,高聚生可下调HLA ABC的表达 ,干扰素 α 2b对HLA ABC抗原表达有较强的上调作用。结论 :高聚生对膀胱癌细胞株无直接的细胞杀伤作用 ,干扰素 α 2b对多个膀胱癌细胞株具有细胞毒作用 ,高聚生抗癌机理与提高肿瘤细胞HLA DR、ABC的表达无关 ,干扰素 α 2b可通过提高肿瘤细胞HLA
OBJECTIVE:To investigate the effects of hight agglutinative staphyeococcin (HAS) and IFN on the cell growth of three types of bladder cancer cells and the expression of HLA DR and HLA ABC. METHODS:Cell culture,MTT assay and FCM were used to dectect the effects of HAS and IFN α 2b on the cell growth of three types of bladder cancer cells and HLA DR, HLA ABC express under the ID 50 culture concentration.RESULTS: HAS emerged a dose dependent manner to promote the cell growth,so that the ID 50 of HAS can not be determined. IFN showed slight suppression on the cell growth and the effects increased along with time.The level of HLA DR expression did not change in three cell lines expose to HAS and IFN α 2b,while the HLA ABC express was down regulate by HAS and up regulate by IFN α 2b.CONCLUSION:HAS has the capacity to promote the cell growth, IFN α 2b show the dose dependent effect on cell growth suppression; The mechanism of antitumor of HAS seems not to relate to the enhanced expression of HLA DR or HLA ABC, Since IFN α 2b is able to increase the expression of HLA ABC expression,enhanced recognization and cytotoxity of CTL to bladder cancer cells.It is may be one of the mechanism of antitumor of IFN α 2b.
