摘要
目的 :探讨外向钾 ( K+ )电流的变化在风湿性心脏病 ( RHD)心房纤颤 ( AF)心房电重构 ( AER)中的作用。方法 :采用酶解法获得人心房肌单个细胞 ,利用膜片钳技术的全细胞记录法 ,记录 RHD AF和窦性心律患者心房肌细胞外向性 K+ 电流的两个主要组分瞬时外向钾电流 ( Ito)和持续外向钾电流 ( IKSUS)。结果 :AF患者心房肌细胞 Ito和 IKSUS在不同除极电压下均较窦性心律患者明显减小 ( P<0 .0 1)。结论 :外向电流减小是复极延缓的因素 ,外向钾电流的减小可能在慢性 AF引起
AIM: To investigate the role of the change in the outward potassium current in the atrial electrical remodeling(AER) caused by atrial fibrillation in rheumatic heart disease. METHODS: Single atrial myocytes were got by enzymatic digestion, and whole cell mode of patch clamp technique was used to measure the densities of transient outward potassium (I to )current and sustained outward potassium current(I KSUS ). RESULTS: The densities of I to and I KSUS in AF patients were significantly lower than those of the patients with sinus rhythm. CONCLUTION: The decrease in outward potassium current is not the main mechanism of AER.
