摘要 目的 :研究细胞周期素A在肾癌组织中的表达及其在肾癌细胞增殖中的作用。方法 :应用免疫组化SABC法检测肾癌及正常肾组织中细胞周期素A及Ki 6 7基因的表达 ,运用统计学方法分析其差异 ,并通过Ki 6 7表达指数评价细胞周期素A在肾癌细胞增殖中的作用。结果 :细胞周期素A在肾癌及正常肾组织中的阳性表达率分别为 12 12 %和 5 0 9% (P <0 0 1)。在肾癌组织中 ,病理分级较高、术后生存时间 <5年者呈高表达 (P <0 0 1) ,不同临床分期及有 (无 )局部淋巴结转移的肿瘤之间表达无显著差异 (P >0 0 5 )。Ki 6 7指数较高者 ,细胞周期素A呈高表达 (P <0 0 1) ,细胞周期素A阳性表达率与Ki 6 7指数显著相关 (r =0 75 7,P <0 0 1)。结论 :细胞周期素A可能参与肾癌的恶性转化 ,但与肿瘤的临床进展过程无关。检测细胞周期素A的水平可反映肾癌细胞的增殖活性 。 Objective: Our aims were to study the expression of cyclin A in renal cell carcinoma (RCC), and to analyze the role of cyclin A in tumor cell proliferation in RCC. Methods: Cyclin A and Ki 67 expressions were studied with immunohistochemical technique in 46 samples of RCC tissues and 13 normal renal tissues. The significance of cyclin A expression was evaluated by Ki 67 index. Results: The positive cell rates of cyclin A in RCC tissues and in normal renal tissues were 12.12% and 5.09%, respectively (P<0 01). Cyclin A expression was related to tumor grade (P<0 01) and prognosis (P<0 01), but not related to tumor stage and metastasis by lymphatics. Cyclin A over expression was observed in the cases with high Ki 67 index. A significant relationship between cyclin A and Ki 67 immunoreactivity was observed (r=0 757,P<0 01). Conclusion: Cyclin A may have a role in carcinogenesis in RCC, but not be involved in tumor progress and metastasis. The immunoreactivity of cyclin A can be used as a clinical prognostic parameter.
