摘要
纳豆激酶是一种新型食源性纤溶酶 ,具有易于提取 ,无任何毒副作用 ,溶栓效果好 ,体内作用时间长等特点。实验以新鲜制备纳豆为原料 ,经生理盐水浸提 ,硫酸铵分级盐析 ,SephadexG -10 0层析获得纯纳豆激酶活性蛋白。实验表明 ,该酶分子量为2 80 0 0Da ,等电点为 9,在 6 0℃以下 ,pH 5~ 12均较稳定 ,其最适反应pH为 8,最适反应温度为 45℃ ,Mg2 + 和Co2 + 离子对酶活有激活作用 ,而Cu2 + 、Zn2 +和Al3 + 等离子对该酶有明显的抑制作用。
Fresh natto was extracted by 0 85% NaCl solution,and then precipitated gradually by(NH 4) 2SO 4. Subseqently nattokinase was obtained by Sephadex G-100 chromatography.The results showed molecular weight of nattokinase was 28,000 Da,and isoelectric point was about 9. It was stable at pH5~12 when the temperature was 60℃.The optimum condition for enzyme was at 45℃ and pH8.Mg 2+ and Co 2+ could activate enzyme activity,Zn 2+ ,Cu 2+ and Al 3+ could inhibate the activity.
