摘要
目的 比较超极化心脏停搏液和去极化心脏停搏液对猪冠状动脉张力的影响。 方法 将新鲜猪心外膜下冠状动脉前降支切成 3段 ,长 3mm,共 2 1条血管环 ,随机分成 3组 ,每组 7条。对照组 :在 37℃有氧条件下浸泡于重碳酸盐缓冲液 (KH) 1小时 ;组 :在同等条件下浸泡于含尼可地尔 (0 .1mmol/ L)的 KH液 1小时 ;组 :在同等条件下浸泡于含氯化钾 (2 0 mm ol/ L )的 KH液 1小时。采用器官槽法 ,检测冠状动脉环静息张力、前列腺素 F2α引发的冠状动脉预收缩强度 ,非受体介导钙离子载体引起的内皮源性血管舒张变化。 结果 与对照组比较 ,组 冠状动脉静息张力降低 (P<0 .0 0 1) ,而非受体介导钙离子载体引起的内皮源性血管舒张变化差别无显著性意义 (P=0 .2 0 3) ;组 静息张力明显升高 (P<0 .0 0 1) ,非受体介导钙离子载体引起的内皮源性血管舒张变化差别具有显著性意义 (P=0 .0 0 3)。前列腺素 F2α引发的冠状动脉预收缩强度 3组间比较差别均无显著性意义 (P=0 .76 0 ,0 .30 5 ,0 .114)。 结论 超极化心脏停搏液能舒张冠状动脉 ,对内皮源性超极化因子 (EDHF)介导血管舒张功能有保护作用 ;去极化心脏停搏液使冠状动脉收缩 ,并有抑制
Objective To compare the effect of hyperpolarizing cardioplegia and depolarizing cardioplegia on the porcine coronary artery tone. Methods Twenty one porcine coronary artery rings in 3 mm long were randomly divided into three groups. Control group ( n =7): Incubation in Krebs Henseleit (KH) at 37℃ for 1 hour with a constant supply of oxygen; groupⅠ( n =7): Incubation in hyperpolarizing cardioplegia (the potassium channel opener, nicorandil 0.1 mmol/L) at the same condition; groupⅡ( n =7): Incubation in depolarizing cardioplegia (high potassium concentration, 20 mmol/L) at the same condition. The basal tone during incubation with potassium or nicorandil and endothelium derived hyperpolarizing factor (EDHF) mediated relaxation (percentage of 30 nmol/L prostaglandin F 2α precontraction) induced by calcium ionophore A23187 in the present of indomethacin (7μmol/L) and N nitro L arginine (300 μmol/L) was measured in the organ chambers. Results Compared with control group, the tone of coronary arteries significantly decreased ( P< 0.001)and EDHF mediated relaxation was unchanged ( P =0 203) in group Ⅰ. In contrast, the tone of coronary arteries significantly increased ( P< 0.001)and EDHF mediated relaxation reduced ( P= 0.003) in group Ⅱ. There are no difference in precontraction among three groups( P= 0.760, 0.305, 0 114). Conclusion Hyperpolarizing cardioplegia has a vasorelaxant effect and can preserve the EDHF related vasorelaxant function;depolarizing cardioplegia contracts the coronary artery and impairs the EDHF related vasorelaxant function.
