摘要
提取迟缓爱德华菌 (Et)外膜蛋白 (OMP) ,经SDS PAGE分析发现 ,在不同培养温度、时间、pH值及不同培养基上OMP图谱无明显差异 ,主条带是 35 0 0 0和 40 0 0 0。但在铁限制环境下 ,增加了高分子区域带 980 0 0 ,930 0 0 ,75 80 0及72 40 0 ,而 45 0 0 0条带变细变浅。用去污剂TritonX 10 0和Sackosyl抽提OMP ,条带基本相似 ,后者主条带更浓密。用SDS抽提OMP ,则仅有 35 0 0 0条带。将OMP样品分别在 37,5 0 ,70 ,90及 10 0℃加热 5min后点样 ,45 0 0 0条带持续出现 ,35 0 0 0在 70℃以上开始出现 ,在 90℃和 10 0℃浓集 ,提示 35 0 0 0条带是热修饰微孔蛋白。
The outer membrane protein(OMP)profiles of Edwardsiella tarda ATCC strain analysed with SDS PAGE showed no obvious difference after the bacteria cultured in different temperatures,time,pH and mediums.The major bands of Et were 35 000 and 45 000.High molecular protein bands such as 98 000,93 000,75 800,72 400 could also be detected if the medium limited iron,while the 45 000 band became slight.Three methods for extracting OMP were used,the major bands were denser by Sackosyl than them by Triton X 100.If treated with SDS, only 35 000 could be found.It appeared to be a heat modifiable protein,as the band could be only detected when the samples heated at the temperatures high than 70 ℃ prior to electrophoresis.
