摘要
目的 观察 GM中毒性 ATI中肾小管上皮细胞凋亡的程度及外源性 EGF对这种细胞凋亡的作用。方法 将雌性 Wistar大鼠分为三组 :1正常对照组 ( NL 组 ,n=7) ;2 GM致 ATI组 ( G组 ,n=2 0 ) ;3 GM+EGF组 ( GE组 ,n=1 9)。分别在 GM或 GM+EGF注射后第 1、4、8、1 2天测定肾小管上皮细胞的 3 HTd R掺入率和血清肌酐 ,并在光镜和电镜下检测肾组织细胞凋亡状况。结果 GM中毒后 ( G组 )动物肾组织细胞凋亡呈双峰现象 ,即在肾组织损伤早期 (第 1天 )和肾组织增殖晚期 (第 8天 )均出现细胞凋亡。而 GM与 EGF共处理后的动物的细胞凋亡在上述两阶段均较 G组动物减少 ,且 3 HTd R掺入率均较 G组动物明显增加。结论 1在 GM中毒性 ATI模型中 ,肾小管上皮细胞凋亡呈双峰现象 ,表明细胞凋亡是早期肾小管损伤的一种表现 ,而肾小管再生阶段出现细胞凋亡可能在维持细胞数平衡中起一定作用。 2外源性 EGF对 GM中毒性
Objective To study the effects of exogenous epidemal growth factor (EGF) on apoptosis in Gentamicin (GM) induced acute tubular injury (ATI) in rats.Methods Female rats were divided into three groups:①Normal rats (Group NL n =7);②GM treated only (Group G, n =20),rats were administered with GM 200mg/kg ×3 days ip;③GM and EGF treated (Group GE, n =19),EGF 20μg was injected subcutaneously after last injection of GM. thymidine incorporation rate ( 3HTdR) of renal tissue and serum creatinine concentration were tested in the day 1?4?8?12 after GM administration,the morphologic changes of apoptotic cells in kidney tissue were examined under light and electronic microscope.Results The count of apoptotic cells (AC) showed two peaks phenomenon:the AC in the kidney tissue of group GE was significantly lower than that of group G at the early phase (lst day) of renal injury and at the late phase (eighth day).Moreover, 3HTdR of renal tissue in group GE was significantly higher than that in group G at several time points after toxic injury.Conclusions ①Apoptosis in mild moderate GM induced ATI appeared at two peaks,the early and late phase;②Administration of exogenous EGF could inhibit apoptosis of renal tubular epithelial cells in GM induced ATI.
