摘要
全面考虑了疏水色谱中蛋白质、固定相和流动相之间的各种相互作用,特别强调了流动相中盐和水的作用。提出了一个能够准确预测较宽浓度范围内蛋白质在疏水色谱中保留值的方程式,以实验数据对此方程进行了验证。结果表明,无论洗脱曲线是否存在极小值,本文所提出的方程式都能予以良好地描述。
Considering all kinds of the interactions among the molecules of protein,stationary phase and mobile phase,especially on the roles of the water and salts in it,we present a retention equation of protein in hydrophobic interaction chromatography in this paper.This equation wasexamined with experimental data and it was found that no matter whether the elution curve of protein appears a concave or not,this equation canaccurately be used to predict the retentions of proteins in hydrophobic interaction chromatography.
