摘要
目的 研究二个针对 c- myc基因的反义脱氧寡核苷酸 (简称反义核酸 )对急性白血病细胞 HL - 6 0的分化及基因表达的作用。 方法 以人工合成二个硫代寡核苷酸作用于 HL - 6 0细胞后 ,采用流式细胞仪检测 c- myc蛋白表达和 HL - 6 0细胞分化抗原 ,RT- PCR方法检测 c- myc基因表达 ,并在光镜下观察以瑞特染色及 NBT染色的HL - 6 0细胞形态。 结果 经反义核酸作用后 ,c- myc蛋白的表达有明显下降 ,与作用浓度与时间相关。急性白血病HL - 6 0细胞出现中 ,晚幼粒细胞增多 ,出现 CD1 5增高 ,HL A- DR及 CD3 4 降低是细胞分化的表现。同时 RT- PCR分析c- myc基因扩增明显减少。 结论 反义核酸对急性白血病细胞 HL - 6 0的诱导分化的同时抑制 c-
Objective\ To study the effects of c\|myc antisense oligodeoxynucleotide(PS\|ODN) on differentiation of HL\|60 cells.\ Methods\ Two sulfur\|modified antisense oligodeoxynucleotides were synthesized and used to act on HL\|60 cells respectively, the level of c\|myc protein and differentiation associated\|antigen of HL\|60 were determined by flow cytometry.\ The expression of c\|myc mRNA was determined by reverse transcription polymerase chain reaction(RT\|PCR).\ Cell morphology was observed under microscope by Wright\|Giemsa and NBT staining.\ Results\ The level of c\|myc protein markedly decreased after treatment by Aspo.\ HL\|60 cells tended to differentiate evidenced by the increased number of myelocyte and metamyelocyte with increasing CD\-\{15\} positive HL\|60 cells and decreased HLA\|DR, CD\-\{34\} positive cells.\ The expression of c\|myc gene at transcription level was markedly decreased by RT\|PCR.\ Conclusion\ c\|myc antisense phosphorothioate oligodeoxynucleotide can induce the differentiation of HL\|60 and inhibit the expression of c\|myc gene.\;
