摘要
目的 :进一步研究SLE病人的ds DNA抗体重链V区基因的组成特点及其VH基因区突变在系统性红斑狼疮狼疮肾炎的发病机理中的作用。方法 :从SLE肾炎病人外周血中分离淋巴细胞制备的噬菌抗体库中克隆出ds DNA单克隆抗体细胞株NYE3,对其IgV基因进行检测和分析。结果 :NYE3单克隆抗体VH在编基因的CDR区出一精氨酸和赖氨酸等带正电荷的氨基酸突变。结论 :SLE病人的ds DNA抗体的VH编码基因的CDR区出现带正电荷的氨基酸突变改变可能与SLE患者肾炎的发病有关。
Objective:To study the charactoristic of VH region of ds DNA antibodies of patients with systemic lupus erythomatosus (SLE) and effects of gene mutation of CDR region of VH gene of the antibody in the pathogenesis of lupus nephritis.Methods:ds DNA monoclonal antibody,NYE3 was cloned from phago display antibody library of patient with SLE and sequenced the VH gene of NYE3.Results:There are many positive amino acid residuals,such as arginine and lysine in CDR3 region of VH region of NYE3 autoantibody.Conclusion:IT suggested that positive amino acid residual mutation of CDR3 region of ds DNA antibodies may play a major role in the pathogenesis of lupus nephritis.
