摘要
利用^(31)P核磁共振技术,对脱氧腺苷-[α-硫代]三磷酸盐的构型进行了研究。在^(31)P-NMR实验中观察到在δ43.1ppm处,有两个三重峰,根据其化学位移和偶合裂分提供的信息,确证了这化合物是5′-脱氧腺苷-[α-硫代]三磷酸盐。同时^(31)P-NMR图谱表明,此化合物是以单一的SP或RP异构体存在。
In the preparation of high specific activity deoxyadenosine 5'-[α-85S] thlotripho-sphate, the structure of deoxyadenosine 5'-α-thiotriphosphate (5'-dATPαS) was determined by 31P-NMR. In the proton coupled 31P NMR spectrum, there was a triplet of doublets (JPα-β=27Hz, JPαH5=6Hz) at 43.1ppm. Therefore this compound was recognized to be 5'-isomer. The 31P-NMR spectrum showed that it exists in either SP or RP configuration.
