摘要
利用腹主动脉缩窄性高血压大鼠模型 ,以平均动脉压 (MAP)和左心室重 /体重比值(LVW /BW )作为指标 ,观察血管紧张素Ⅱ (AngⅡ )受体拮抗剂芦沙坦 (Losatan ,Los)在压力超负荷高血压和心肌肥大反应过程中的作用 .结果发现 :1.腹主动脉缩窄 (COA)大鼠术后第 1、4、8、12周MAP明显增高 ,并呈时间依赖性 ;COA组大鼠与假手术 (Sham)组大鼠各时间点的MAP比较均显著升高 .2 .COA大鼠术后第 1、4、8、12周LVW /BW明显增高 ,并呈时间依赖性 ;COA组大鼠与Sham组大鼠各时间点的LVW/BW比较均显著升高 .3.Los明显阻断COA大鼠MAP和LVW /BW的增高效应 ,与单纯COA组大鼠相比有极显著性差异 ;但是 ,Los对Sham组大鼠的MAP和LVW/BW无明显影响 .以上结果表明 :腹主动脉缩窄可作为高血压和心肌肥大反应的动物模型 ;AngⅡ参与诱发压力超负荷高血压和心肌肥大反应的发生 .
Using the model of rat with the hypertension of abdominal aorta coarctation(COA),and taking the mean arterial pressure(MAP) and the ratio of left ventricular weigh to body weight(LVW/BW) as an index,the authors observed the effects of Losatan(LOS)-angiotensin Ⅱ 1 st type receptor inhibitor-on the rats' pressure overload hypertension and cardiac hypertrophy. It was found that:the MAP and LVW/BW significantly increased at the 1 st,4 th,8 th,and 12 th weeks after rats' COA operation,and had a relevance with time; while compared with the rats' MAP and LVW/BW of Sham group,both the rats'MAP and LVW/BW in COA group obviously increased at every time unit.On the other hand,Los could obviously reduced COA rats' MAP and LVW/BW,but it didn't obviously affect on the rats' MAP and LVW/BW in Sham group.The above results showed that COA rats could be used as an animal model of hypertension and cardiac hypertrophy; and that Ang Ⅱ had participated in the reactive process of inducing the rats' pressure overload hypertension and cardiac hypertrophy.
